C/I: Hypersens; Co-admin. with nitric
oxide donors (such as amyl nitrite) or
nitrates in any form due to the
hypotensive effects of nitrates.
Co-admin. of PDE5 inhib., includ.
sildenafil, with guanylate cyclase
stimulat; Comb. with the most potent of
the CYP3A4 inhib. (eg, ketoconazole,
itraconazole, ritonavir).
Pts. who have loss of vision in one eye
because of non-arteritic anterior
ischaem. optic neuropathy (NAION),
regardless of whether this episode was
in connect. or not with prev. PDE5 inhib.
expos; Sev. hep. impair; Recent history
of stroke or MI; Severe hypotens. (blood
pres.< 90/50 mmHg) at initiat.

Phosphodiesterase-5 Enzyme Inhibitor. Sildenafil 25 mg.
Susp. for oral spray 20 ml. Each actuat. delivers 0.5 ml susp., contain. 12.5 mg sildenafil
Dose is 2 mL (4 act.), equiv. to 50 mg of sildenafil taken as needed, approx. one hour before sexual activ. Based on effic. and tolerab., dose may be incr. to 4 mL (8 act.), equiv. to 100 mg, or decr. to 1 mL (2 act.), equiv. to 25 mg of sildenafil. Max. dose is 4 mL (8 act.), equiv. to 100 mg of sildenafil. Max. dosing freq. is 1/d.
In adult men with erectile dysfunction.
C/I: Hypersens. Co-administr. with nitric oxide donors (such as amyl nitrite) or nitrates in any form. Co-administr. with guanylate cyclase stimul., such as riociguat, as it may lead to symptom. Hypotension. Not  to be used in men for whom sexual act. is inadvisable (e.g. pts. with sev. CV disord. such as unstable angina or sev. HF). Contraindic. in ptts. who have loss of vision in one eye due to non-arteritic anterior ischaemic optic neuropathy (NAION). Safety has not been studied in the follow. sub-groups of pts. and use is contraindic. in these pts: sev. hepatic impairm., hypotension (blood pressure <90/50 mmHg), recent hist. of stroke or MI and known heredit. degenerat. retinal disord. such as retinitis pigmentosa.

To be taken once dly. 50 mg as needed
approx. 1 h bef. sexual act.; may be
incr. to 100 mg or decreas. to 25 mg.

after meal.
Impair. renal funct.: mild-mod.: no
dosage adjust. req.; Sev. renal impair.:
25 mg should be consid. Impair. hepatic
funct.: A dose of 25 mg should be
consid.
Elderly: Dosage adjust. is required.

C/I: Known hypersens. to product; pts.
concur. using nitric oxide donors (such
as amyl nitrite), organic nitrates, or
organic nitrites in any form either
regularly or intermittently; under 18
years; sev. cardiovascular or sev. liver
disease; recent history of MI ,stroke, or
pts. who suffer from hypotension; pts.
who have loss of vision in one eye
because of non-arteritic anterior
ischemic optic neuropathy (NAION);
hereditary degener. retinal dis. such as
retinitis pigmentosa; not intended to be
used in women.
