Gamma-Aminobutyric Acid Analog. Pregabalin 25 mg, 50 mg, 75 mg, 100 mg, 150mg, 200 mg, 225 mg, 300 mg. CAPS: 56 x 25 mg, 50 mg, 75 mg, 100 mg, 150mg, 200 mg, 225 mg, 300 mg. The dose range is 150- 600 mg/d given in either two or three divid. doses.
Neuropath. pain: Init. 150 mg per day given as two/ three divid. doses. Based on individ. pt. response and tolerab., the dose may be incr. to 300 mg/ d after an interv.of 3 - 7 days, and if needed, to a max. dose of 600 mg/ d after an addit. 7-day interval.
Fibromyalgia: The usual dose 300 -450 mg/d given in two divid. doses. Some pts. may derive addit. benefit at 600 mg/ d. Dosing should begin at 75mg ×2/d (150 mg/d) and may be incr. to 150 mg ×2/d t (300 mg/d) within 1 wk. based on effic. and tolerab. Pts. who do not experience suffic. benefit with 300 mg/d may be further incr. to 225 mg ×2/d (450 mg/d). If needed, in some pts., based on individ. response and tolerab., the dose may be incr. to max. dosage of 600 mg/d after an addit. wk.
Discont. of pregabalin: It is recommended that the discont. of the drug will be done gradual. over a minim. 1 wk. independent of the indicat.
Renal impair.: For pts. receiving haemodialysis, the pregabalin dly. dose should be adjust. based on ren. funct. In addit. to the dly. dose, a supplem. dose should be given immediate. follow. every 4-h haemodialysis tmt.
Hep. impair.: No dosage adjust. is needed. See lit.
