Phosphatidylinositol-3-Kinase (PI3K) Inhibitors. Alpelisib 50,100,200 mg. F.C.TABS.:28. The recommend. dose is 300 mg, 2 tabs (150mg) ×1/d y, with food. until dis. progress. or unacceptab. toxic. occurs. Pts. should take their dose of approx. at the same time each day. tabs. should not be chewed, crushed or split prior to swallow. If a dose is missed, it can be taken with food within 9 hrs. after the time it is usually taken. After more than 9 hrs., skip the dose for that day. The next day, take the dose should be taken at usual time.
If the pt. vomits after taking the dose, addit. dose should not be taken and
the admin. may be resumed the dosing schedule the next day at the usual time.
When given concom. with fulvesstrant, the recommended dose of fulvestrant is 500 mg admin. on Days 1, 15, and 29, and once mnthly. thereafter. See lit.
Indic. in comb. with fulvestrant for the tmt. of postmenopaus. women, and men, with hormone receptor (HR)-posit., human epidermal growth factor receptor 2 (HER2)-neg., PIK3CA- mutated, advanced or metastat. breast canc. follow. progress. on or after an endocrine-based regimen.
