    Dosage

    30-300 mg daily in divided doses according to individual requirement, 75 mg/day is recommended.


    Indications

    Anxiety, depression, mixed anxiety depressive states.


    Contra-Indications

    Known hypersensitivity to tricyclic antidepressives, concomitant administration with MAO inhibitors, acute recover phase following myocardiac infarction.


    Special Precautions

    Pregnancy and lactation, children under 12 years, manic depressive, schizophrenic and potentially suicidal patients. Patients with cardiovascular disorders, patients with a history of urinary retention, angle-closure glaucoma, or increased intraocular pressure. Close supervision and careful adjustment of dosage are required when this drug is given with anticholinergic or sympathomimetic drugs. Caution in hyperthyroid patients, or those receiving thyroid medicdbation, patients with a history of seizure disorders, concurrent use with monoamine oxidase inhibitors.


    Side Effects

    Dryness of the mouth, blurred vision, disturbances of accomodation, constipation, drowsiness, confusional states, hallucinations, disturbed concentration, peripheral neuropathy, fine tremor, seizures, extrapyramidal symptoms. Paradoxical reactions include anxiety and insomnia. Allergic skin reactions and photosensitivity, rare cases of agranulocytosis, leukopenia, leukocytosis, eosinophilia and thrombocytopenia have been reported. Heartburn, nausea, vomiting, unpleasant taste, diarrhea anorexia. Cholestatic jaundice, testicular swelling and gynecomastia in the male, breast enlargemnt and galactorrhea in the female, increased or decreased libido, alopecia, edema, weight gain or loss, urinary frequency, increased perspiration, increases or decreases in blood glucose levels.


    Drug interactions

    MAOIs, directly acting sympathomimetics, antihypertensives, phenothiazines, anticholinergics, anticonvulsants. Alcohol, antihistamines, CNS depressants, benzodiazepines, opiate analgesics, cimetidine, phenytoin. Barbiturates, disulfiram, sublingual glyceryl trinitrate, thyroxine, acetazolamide. Zopiclone, high fibre diet, neuroleptics, SSRIs.
